醋柳黄酮对高血压大鼠主动脉环舒缩反应的影响及机制

【摘要】ObjectiveToexplorethemechanismoftotalflavonesofhippophaerhamnoides(TFHintreatingabdominalaorticcoarctation-inducedhypertensioninratsanddetectthecontractilerelaxantresponseofaortarings.MethodsExperimentalhypertensiveratmodelswereestablishedbypartiallybandingabdominalaorticarteryandwererandomlydividedintohypertensivemodelgroup(n=10,potablewater10mL/kg,TFHgroup(n=10,30mg/kg,Captoprill(CPgroup(n=10,100mg/kgandTFH+CPgroup(n=10,TFH20mg/kg+Captoprill60mg/kgfor6weekswhilesham-operatingascontrolgroup(n=10,potablewater10mL/kg.Then,detectthecontractilerelaxantresponseofaorticringsandserumNitricOxide(NOlevels.ResultsComparedwithmodelgroup,bothTFHgroupandcaptoprilgroupobvioureaslyincreasedthelevelofserumNO(P<0.05,especiallyTFHgroup.AndtherewassynergeticeffectofTFHandCaptoprill.Thevasodilationeffectsof3treatedgroupsincreasedinvaryingdegreecomparedwiththemodelgroup(P<0.05whiletherewerenosignificantdifferencesamongthosegroups.PretreatedwithL-NAME(aninhibitorofNOS,therelaxantresponseinducedbyAcetylcholine(AchofTFH+CPgroupwassignificantlyhigherthanthatofthemodelgroup(P<0.05.Thecontractileresponseofinducedbynoradrenalinibitartas(NEofTFH+CPgroupwasobviouslylowerthanthatofthemodelgroup(P<0.05.InlowconcentrationofNE,thecontractileresponseofTFHandCP
groupsdecreasedinvaryingdegreecomparedwiththemodelgroup(P<0.05.ConclusionTFHlong-termusecanimprovetheen-dothelialfunctionofhypertensiverats,reducevascularsensitivitytothevasoconstrictorsubstance.TheseeffectsofTFHaremediatedbyimprovingendothelialfunctionandincreasingthelevelofserumendothelium-derivedNO.TheeffectsofTFHareequaltoCaptoprill,andthereissynergeticeffectofthem.%目的探讨腹主动脉缩窄高血压大鼠服用醋柳黄酮(TFH治疗后胸主动脉环的舒缩反应及机制.方法采用腹主动脉缩窄法制备高血压大鼠模型,随机分为4组,每组10只:模型组饮用水10mL/kg;TFH组TFH30mg/kg;卡托普利组(CP组CP100mg/kg;TFH+CP组TFH20mg/kg和CP60mg/kg,连续灌胃给药,每日1次,10只假手术组作为对照组.用药6周后,进行主动脉环舒缩反应测定,检测血一氧化氮(NO浓度.结果TFH、CP能明显升高高血压大鼠的血NO水平(P<0.05,以TFH更明显,且TFH和CP有协同作用.用药组大鼠内皮依赖性舒张敏感性较模型组显著增加(P<0.05,用药组之间差异无统计学意义.NO合成酶抑制剂左旋硝基精氨酸甲酯(L-NAME预处理后,对乙酰胆碱(Ach诱发的舒张反应,TFH+CP组明显高于模型组(P<0.05.对重酒石酸去甲肾上腺素(NE诱发的收缩反应,TFH+CP组显著低于模型组(P<0.05.TFH组和CP组在NE浓度较低时收缩反应低于模型组(P<0.05.结论TFH长期服用可以改善高血压大鼠内皮舒张功能,降低血管对缩血管物质的敏感性,其机制与保护血管内皮和刺激内皮源性NO增加有关;TFH的舒血管作用与卡托普利等效,两药联用具有协同作用.【期刊名称】《重庆医学》【年(卷,期】2011(040029【总页数】4页(P2955-2958

醋柳黄酮(totalflavonesofhippophaerhamnoidesL,TFH是一种从野生胡颓子科植物醋柳(俗称沙棘的果实中提取出的一种有效成分。TFH含有7种单体成分,其中以槲皮素(quercetin,Que及异鼠李素(isorhamnetin,Iso含量较高。TFH具有重要的心血管保护作用,包括抗氧化、扩血管、降压、抗凝、抑制心肌肥厚和保护内皮功能等[1]。血管内皮细胞可合成和分泌一氧化氮(NO,NO具有强有力的扩张血管、降低血压、抑制血小板黏附和聚集等作用,是已知的具有抗高血压作用的保护性因子。本实验用TFH灌胃治疗腹主动脉缩窄高血压大鼠6
